Glad you managed to get that working in the end.
You want Xplorer 360 Extreme2 to copy his content off the 20gig drive and Xplorer 360 250gb Edition to copy content to the new 250gig drive.
Before copying any content you might want to have a look at this post, it details how to work around a problem files you inject end up in the wrong place. Remember to run Xplorer 360 with admin rights.
I believe you can disable UAC on Vista / Windows 7 then reboot and Xplorer 360 will allow you to drag and drop which means you will not have the above problem. If your on XP it should just work as long as your user account has admin rights.
On your 20gig drive delete anything on it you dont need, such as installed games, demos or any videos you dont need. The 250gig edition is very slow at injecting content, however it will get there in the end.
If you have a USB drive you could format it for use in the USB (an option in the memory management screen), copy all your content on to that, take the 20gb drive out, put the 250gig drive in then copy the content to the 250gig drive. Ofcourse that depends how big of a usb drive you have.
Also don't forget to restore partition 2, that is needed to play Xbox 1 games.
